New opportunities for radio-immunotherapy in head and neck cancer

The effects of combining immunotherapy with radiotherapy are dependent on radiation dose, fractionation and irradiated volumes in preclinical models. In addition to immune checkpoint inhibition several other immunotherapy approaches are on the horizon. HPV positive head and neck cancers have a distinct immune microenvironment with immune cell infiltration and PD-L1 expression and might be a specially interesting subgroup of patients for immunotherapy approaches. Published results of clinical trials have shown tolerability of combined treatment regimens, but have also shown limited benefit in some randomized settings.
